    A B23-interacting sequence as a tool to visualize protein interactions in a cellular context.

    International audienceWe report the characterization of a nucleolar localization sequence (NoLS) that targets the green fluorescent protein (GFP) into the granular component (GC) of nucleoli. This NoLS interacts in vitro specifically and directly with the major nucleolar protein B23 and more precisely with the region of B23 including the two acidic stretches. The affinity of NoLS for B23 is stronger than that of the HIV-1 Rev protein in vitro. Moreover, B23-NoLS interaction also occurs in vivo. Indeed, (1) NoLS confers on the GFP the behavior of B23 throughout the cell cycle, (2) the GFP-NoLS fusion and B23 remain colocalized after drug treatments, (3) a selective delocalization of B23 from nucleoli to nucleoplasm induces a concomitent delocalization of the GFP-NoLS fusion, and (4) the fusion of NoLS to fibrillarin makes it possible to colocalize fibrillarin and B23. Interestingly, by fusing NoLS to fibrillarin, both fibrillarin and the fibrillarin partner Nop56 are mislocalized in the GC of nucleoli. Similarly, by fusing the NoLS to MafG, part of the nuclear transcription factor NF-E2 composed of both MafG and p45 NF-E2, NF-E2 is redirected from the nucleoplasm to the nucleoli. Thus, we propose that the NoLS may be used as a tool to visualize and prove protein interactions in a cellular context

    The pilot dimension method: Reconciling Steering and Conformity in Workshops

    International audienceIn machining workshops, workpieces are produced according to dimensions known as manufacturing dimensions. For the same workpiece and the same manufacturing plan, several sets of manufacturing dimensions can be used but none satisfy simultaneously the two main missions workshops need to fulfil: (a) Ensuring conformity of products to their design dimension tolerances (also called blueprint tolerances) and (b) steering machines in order to compensate for tool wear. The set of manufacturing dimensions obtained from the design dimensions using the minimal chain of dimensions method is optimal for a conformity check of workpieces but is practically unusable for steering machines because of the complexity of its relationships toward the tool correctors and tools dimensions. The pilot dimensions method consists in, on the one hand, identifying and representing these tool correctors and these tool/program dimensions on the production drawings (besides the manufacturing dimensions) and, on other the other hand, determining their correction values through a mathematical set of relations after having measured the manufacturing dimensions on a workpiece. Doing so will strongly reduce adjustment time, reduce the number of workpieces used for adjustments and greatly enhance the quality of workpiece batches

    Recent advances on the utilization of layered double hydroxides (LDHs) and related heterogeneous catalysts in a lignocellulosic-feedstock biorefinery scheme

    Layered double hydroxides (LDHs) and derived materials have been widely used as heterogeneous catalysts for different types of reactions either in gas or in liquid phase. Among these processes, the valorization/upgrading of lignocellulosic biomass and derived molecules have attracted enormous attention because it constitutes a pivotal axis in the transition from an economic model based on fossil resources to one based on renewable biomass resources with preference for biomass waste streams. Proof of this is the increasing amount of literature reports regarding the rational design and implementation of LDHs and related materials in catalytic processes such as: depolymerization, hydrogenation, selective oxidations, and C-C coupling reactions, among others, where biomass-derived compounds are used. The major aim of this contribution is to situate the most recent advances on the implementation of these types of catalysts into a lignocellulosic-feedstock biorefinery scheme, highlighting the versatility of LDHs and derived materials as multifunctional, tunable, cheap and easy to produce heterogeneous catalysts

    First dating results for the Middle Pleistocene industries (Acheulean – Early Middle Palaeolithic) in the Pyrenees – Garonne region: a multi methods geochronological approach (TL, OSL and TT-OSL) of the Duclos and Romentères sites

    The recent development work of the A65 highway has given the opportunity to broaden the corpus of Lower and Middle Palaeolithic open-air sites known in the southern Aquitaine basin. The sites of Duclos (Auriac, Pyrénées-Orientales) and Romentères (Le Vignau, Landes), discovered in this context, have yielded an abundant lithic record which has been attributed to the Acheulean of Iberian type for the earlier and, essentially, to the Early Middle Palaeolithic for the latter. The archaeological levels are intercalated in sequences that comprise aeolian silts and colluvial units separated by interglacial luvisols. The pedostratigraphic context of the two sites has provided a rare opportunity to propose a reliable chronostratigraphic framework in this area. To achieve this objective, a study combining several methods of luminescence dating was conducted to complete the geomorphological data. Optically Stimulated Luminescence (OSL) and Thermically Transferred OSL (TT-OSL) dating methods were applied in order to determine the ages of sedimentary quartz grains and thus, of the sampled deposits. In parallel, heated flint and quartzite samples were dated by thermoluminescence (TL). The Pyrenees-Garonne Acheulean industry of Duclos has been attributed to the boundary between Marine Isotopic Stages (MIS) 7 and 6 while the human occupations of Romentères date from MIS 6 for the most recent series (Early Middle Palaeolithic) and from MIS 9 and 8 for the older

    Sperm Motility and Lactate production at different sperm concentrations

    Lactate production is associated with total spermatozoa concentration. It negatively affects preservation of total and progressive motility, showing an effect of by-products of anaerobic metabolism on long-term storage. Moreover, our data show that non-progressive motile spermatozoa are highly associated to lactate concentration, and thus, anaerobic glycolysis. More studies are required to determine relative contributions of aerobiosis and anaerobiosis to spermatozoa motility under different storage conditions.Peer reviewe

    Glucose use and lactate production by equine fresh semen in human and equine extender

    This study shows that this human semen extender doesn’t support equine semen preservation. Sperm cells’ glucose consumption and lactate production seem to be negligible, as these parameters were not affected by sperm concentrations in our study. Our results suggest that spermatozoa are able to cleave complex carbohydrates as glucose concentration in INRA96 increased over time

    Aide au tolérancement tridimensionnel (modèle des domaines)

    Face à la demande de plus en plus exigeante en terme de qualité et de coût de fabrication des produits manufacturés, la qualification et quantification optimal des défauts acceptables est primordial. Le tolérancement est le moyen de communication permettant de définir les variations géométriques autorisé entre les différents corps de métier intervenant au cours du cycle de fabrication du produit. Un tolérancement optimal est le juste compromis entre coût de fabrication et qualité du produit final. Le tolérancement repose sur 3 problématiques majeures: la spécification (normalisation d'un langage complet et univoque), la synthèse et l'analyse de tolérances. Nous proposons dans ce document de nouvelles méthodes d'analyse et de synthèse du tolérancement tridimensionnel. Ces méthodes se basent sur une modélisation de la géométrie à l'aide de l'outil domaine jeux et écarts développé au laboratoire. La première étape consiste à déterminer les différentes topologies composant un mécanisme tridimensionnel. Pour chacune de ces topologies est définie une méthode de résolution des problématiques de tolérancement. Au pire des cas, les conditions de respect des exigences fonctionnelles se traduisent par des conditions d'existence et d'inclusions sur les domaines. Ces équations de domaines peuvent ensuite être traduites sous forme de système d'inéquations scalaires. L'analyse statistique s'appuie sur des tirages de type Monte-Carlo. Les variables aléatoires sont les composantes de petits déplacements des torseur écarts défini à l'intérieur de leur zone de tolérance (modélisée par un domaine écarts) et les dimensions géométriques fixant l'étendue des jeux (taille du domaine jeux associé). A l'issue des simulations statistiques, il est possible d'estimer le risque de non-qualité et les jeux résiduels en fonction du tolérancement défini. Le développement d'une nouvelle représentation des domaines jeux et écarts plus adapté, permet de simplifier les calculs relatifs aux problématiques de tolérancement. Le traitement local de chaque topologie élémentaire de mécanisme permet d'effectuer le traitement global des mécanismes tridimensionnels complexes avec prise en compte des jeux.As far as the demand in quality and cost of manufacturing increase, the optimal qualification and quantification of acceptable defects is essential. Tolerancing is the means of communication between all actors of manufacturing. An optimal tolerancing is the right compromise between manufacturing cost and quality of the final product. Tolerancing is based on three major issues: The specification (standardization of a complete and unequivocal language), synthesis and analysis of the tolerancing. We suggest in this thesis some new analysis and synthesis of the three-dimensional tolerancing. These methods are based on a geometric model define by the deviations and clearances domains developed on the laboratory. The first step consists in determining the elementary topology that composes a three-dimensional mechanism. For each kind of these topologies one resolution method is defined. In worst case, the condition of functional requirement respect is traduced by existence and inclusions conditions on the domains. Then these domains equations can be translated in inequalities system of scalar. The statistical analysis uses the Monte-Carlo simulation. The random variables are the small displacements components of the deviation torsor which is defined inside its tolerance area (model by a deviations domain) and the geometrics dimensions which set the extent of clearance (size of the clearance domain). Thanks to statistical simulation, it is possible to estimate the non-quality rate in regards to the defined tolerancing. The development of a new representation of clearances and deviations domains most suitable, allows us to simplify the calculation for tolerancing problems.     Evolution monitoring of a solution-mining cavern in salt : identifying and analysing early-warning signals prior to collapse

    International audienceRisk management of underground cavities requires a good working knowledge of accidental phenomena like subsidence or large-scale collapse. This was the context when the opportunity was taken to instrument a large size in use saline cavern, so as to test various auscultation techniques available under controlled conditions. A microseismic monitoring network coupled to a surface measurement system was installed to improve our knowledge of the mechanisms that initiate and govern the evolution of the cavern up to its collapse. After a stationary period combined with partial depressurization tests conducted in 2005 and 2007, the cavern appears to have entered into its final evolution phase, and this probably since early 2008. This results in continuous and highly sustained microseismic activity as well as the occurrence of a number of microseismic episodes localized around the cavern roof. The localization of the microseismic events, for some of these episodes, is closely correlated to the quasi-dynamic brine pressure variations and to the evolutions of the roof depth measured at observation boreholes. The microseismic activity turns out to be more precise when it comes to the evolution affecting the mine cavern than the movement measurements taken on the surface or sub-surface

    (Shortened) [...] After recalling the basic features of the "fireshell model", we emphasize the following novel results: 1) the interpretation of the X-ray flares in GRB afterglows as due to the interaction of the optically thin fireshell with isolated clouds in the CircumBurst Medium (CBM); 2) an interpretation as "fake - disguised" short GRBs of the GRBs belonging to the class identified by Norris & Bonnell [...] consistent with an origin from the final coalescence of a binary system in the halo of their host galaxies with particularly low CBM density [...]; 3) the first attempt to study a genuine short GRB with the analysis of GRB 050509B, that reveals indeed still an open question; 4) the interpretation of the GRB-SN association in the case of GRB 060218 via the "induced gravitational collapse" process; 5) a first attempt to understand the nature of the "Amati relation", a phenomenological correlation between the isotropic-equivalent radiated energy of the prompt emission E_{iso} with the cosmological rest-frame \nu F_{\nu} spectrum peak energy E_{p,i}. In addition, recent progress on the thermalization of the electron-positron plasma close to their formation phase, as well as the structure of the electrodynamics of Kerr-Newman Black Holes are presented. An outlook for possible explanation of high-energy phenomena in GRBs to be expected from the AGILE and the Fermi satellites are discussed. As an example of high energy process, the work by Enrico Fermi dealing with ultrarelativistic collisions is examined. It is clear that all the GRB physics points to the existence of overcritical electrodynamical fields. In this sense we present some progresses on a unified approach to heavy nuclei and neutron stars cores, which leads to the existence of overcritical fields under the neutron star crust.Comment: 68 pages, 50 figures, in the Proceedings of the XIII Brazilian School on Cosmology and Gravitation, M.     Investigar, ensinar e aprender são actividades que podem estar presentes, de forma articulada, no ensino-aprendizagem da Matemática e na actividade profissional do professor. Para isso, é necessário conceber tarefas que possam ser o ponto de partida para investigações e explorações matemáticas dos alunos e discutir o modo como podem ser trabalhadas na sala de aula. Recorrendo a exemplos de actividades realizadas por professores de Matemática portugueses, analiso a actividade de aprendizagem suscitada por tarefas deste tipo e discuto as respectivas potencialidades. Finalmente, refiro as condições respeitantes à cultura profissional dos professores que podem favorecer uma actividade de investigação sobre a sua própria profissional, com relevo para a colaboração e a dimensão associativa.Researching, teaching and learning are activities that may be present, in coordination, teaching and learning of mathematics in the work of the teacher. Therefore, it is necessary to design tasks that may be the starting point for mathematical investigations and explorations of the students and discuss how they can be worked in the classroom. Drawing on examples of activities carried out by Portuguese teachers of mathematics by examining the activity of learning raised by such tasks and discuss their potencial. Finally, I refer to conditions relating to the professional culture of teachers who can promote the research activity on their own training, with an emphasis on collaboration and associative dimension
